OBJECTIVE: To determine the frequency of and risk factors for major adverse drug reactions (MADRs) associated with anti-tuberculosis treatment at a tuberculosis (TB) referral hospital in the Republic of Korea.METHODS: Data from an ongoing natural history cohort study were analyzed for permanent regimen changes due to adverse drug reactions and confirmed by chart review.RESULTS: Among 655 subjects, there were 132 MADRs in 112 (17%) subjects. The most common MADRs were gastrointestinal (n = 53), musculoskeletal (n = 22), psychiatric (n = 10), visual (n = 9) and peripheral neuropathic (n = 8). MADRs were more frequent in subjects being treated with second-line regimens (16%) compared to first-line regimens (2.5%). Drugs frequently associated with MADRs were amikacin (3/10, 30%), linezolid (8/29, 28%), para-aminosalicylic acid (47/192, 24%), pyrazinamide (31/528, 5.8%), macrolides (2/44, 4.5%) and cycloserine (12/272, 4.4%). Fluoroquinolones accounted for a single MADR (1/377, 0.003%), despite widespread usage. In multivariate analysis, infection with multi- or extensively drug-resistant disease and previous history of anti-tuberculosis treatment were risk factors for MADR, with adjusted hazard ratios of respectively 2.2 (P = 0.02) and 1.6 (P = 0.04).CONCLUSION: MADRs are common during antituberculosis chemotherapy in this population, occurring in more than one in six subjects. New and less toxic agents to treat drug-resistant TB are urgently needed.

The aim of this study was to assess the HIV/AIDS-related knowledge among junior high-school students in Shanghai, China, and the factors influencing this knowledge. A cross-sectional study was conducted in 20 middle schools of two districts by a cluster-stratified selection procedure in Shanghai, China. The 2432 sampled students, aged from 11.1 to 16.7 years, completed a self-administered questionnaire of HIV/AIDS prevention. The results showed that the overall correct rate of HIV/AIDS-related knowledge was 62%. Using multivariable logistic regression analysis, we found that the main factors influencing HIV/AIDS-related knowledge among junior high school students were the type of school (odds ratio [OR]= 1.641), age (OR = 1.727), whether the student was a single child in the family (OR = 1.389), whether the student had previous HIV/AIDS-related education experience (OR = 2.003) and whether the student had ever discussed HIV/AIDS with their parents (OR = 1.282). The results indicate that HIV/AIDS-related knowledge among Shanghai junior high school students is not high enough, and more attention needs to be paid to enhance HIV/AIDS-related knowledge, especially among younger students from common type schools without HIV/AIDS-related education experience. We encourage Chinese parents to get involved in their children's HIV/AIDS prevention education.

Foxtail millet (Setaria italica), a member of the Poaceae grass family, is an important food and fodder crop in arid regions and has potential for use as a C(4) biofuel. It is a model system for other biofuel grasses, including switchgrass and pearl millet. We produced a draft genome (∼423 Mb) anchored onto nine chromosomes and annotated 38,801 genes. Key chromosome reshuffling events were detected through collinearity identification between foxtail millet, rice and sorghum including two reshuffling events fusing rice chromosomes 7 and 9, 3 and 10 to foxtail millet chromosomes 2 and 9, respectively, that occurred after the divergence of foxtail millet and rice, and a single reshuffling event fusing rice chromosome 5 and 12 to foxtail millet chromosome 3 that occurred after the divergence of millet and sorghum. Rearrangements in the C(4) photosynthesis pathway were also identified.

Electrically pumped lasing from Germanium-on-Silicon pnn heterojunction diode structures is demonstrated. Room temperature multimode laser with 1mW output power is measured. Phosphorous doping in Germanium at a concentration over 4x10<sup>19</sup>cm<sup>-3</sup> is achieved. A Germanium gain spectrum of nearly 200nm is observed.

The glomeruli are the first synaptic relay on the olfactory pathway and play a basic role in smell perception. Glomerular degeneration occurs in humans with age and in Alzheimer's disease (AD). The glomeruli heavily express β-amyloid precursor protein (APP), β -secretase (BACE1) and γ-secretase complex. However, extracellular Aβ deposition occurs fairly rarely at this location in postmortem pathological studies. We sought to explore age-related glomerular changes that might link to alteration in amyloidogenic proteins and/or plaque pathogenesis in transgenic models of AD and humans. Focally increased BACE1 immunoreactivity (IR) in the glomerular layer was identified in several transgenic models, and characterized systematically in transgenic mice harboring five familiar AD-related mutations (5XFAD). These elements were co-labeled with antibodies against APP N-terminal (22C11) and Aβ N-terminal (3D6, 6E10) and mid-sequence (4G8). They were not co-labeled with two A β C-terminal antibodies (Ter40, Ter42), nor associated with extracellular amyloidosis. These profiles were further characterized to be most likely abnormal olfactory nerve terminals. Reduced glomerular area was detected in 6-12 month-old 5XFAD mice relative to non-transgenic controls, and in aged humans relative to young/adult controls, more robust in AD than aged subjects without cerebral amyloid and tau pathology. The results suggest that olfactory nerve terminals may undergo age-related dystrophic and degenerative changes in AD model mice and humans, which are associated with increased labeling for amyloidogenic proteins but not local extracellular Aβ deposition. The identified axon terminal pathology might affect neuronal signal transmission and integration at the first olfactory synaptic relay.

Microcystins (MCs) are highly liver-specific and evidenced as a liver tumor promoter. Oxidative stress is one of the most important toxicity mechanisms of MCs, which is tightly related to oxygen concentration. The effects of MCs on animals and cell lines in normoxia and the mechanisms have been well studied, but such effects in different oxygen conditions were still unclear. The aim of the present study was to explore the cellular response of the human hepatocellular carcinoma cell line (HepG2) to MC-LR exposure under hypoxic (1% O(2)) and normoxic (21% O(2)) conditions. We examined cell viability, mitochondrial membrane potential (MMP), superoxide dismutase (SOD) activity and gene expression posttoxin exposure. Cell viability was increased by MC-LR in normoxia although decreased in hypoxia. MC-LR markedly induced MMP loss under hypoxic condition but only slightly MMP loss under normoxic condition. SOD activity was significantly induced by MC-LR in hypoxia, indicating prolonged oxidative stress. Inhibitory apoptosis protein (c-IAP2) was significantly up-regulated by MC-LR under normoxic condition, suggesting that c-IAP2 played an important role in the promotion of cell proliferation by MC-LR. These results indicate that MC-LR promotes cell proliferation under normoxic condition whereas induces cell apoptosis under hypoxic condition. Copyright © 2012 John Wiley & Sons, Ltd.

PURPOSE: The study aims to find candidate probes of fluorescence in situ hybridization (FISH) for detection of lung cancer with bronchial brushings and to evaluate whether the accuracy of diagnosing lung cancer by cytological deviant and genetic abnormalities is greater than that of cytology alone. METHODS: Centromeric enumeration probes (CEPs) for chromosomes 2, 3, 6, 7, 8, 9, 11, 12, and 17 were analyzed using FISH in 74 surgical resection tissues, 32 operative margin tissues without tumor involvement of lung cancer, and 174 bronchial brushings. RESULTS: The aneuploidy rates of the tested probes were 61.7, 89.1, 80.0, 92.7, 65.0, 70.4, 66.7, 71.8, 68.9 % in tumor tissues, and 29.3, 58.9, 33.3, 69.6, 67.0, 40.3, 38.0, 49.3, 35.1 % in bronchial brushings. The combination of cytology and FISH using the three-probe set for chromosomes 3+7+8 significantly improved the sensitivity of bronchial brushing examination for lung cancer detection (P = 0.00003), especially squamous cell carcinoma (SCC), which increased from 78.0 to 98.2 %. The specificity of the 3+7+8 probe set was 94.6 %. Moreover, a high aneuploidy rate of the probe set in bronchial brushings was detected more often in SCCs (P = 0.029) and late-stage non-small-cell lung cancer (NSCLC)(P = 0.044). Kaplan-Meier curves indicated that adenocarcinoma (ADC) patients with high aneuploidy rate of CEP3 in tissue samples exhibited poorer overall survival (P = 0.016). CONCLUSIONS: FISH performed on cytology preparations is useful for confirmation of cancer diagnosis. The three-probe set, 3+7+8, has potential value for the detection of SCCs in bronchial brushings.

The propagation of a partially coherent hollow vortex Gaussian beam through a paraxial ABCD optical system in turbulent atmosphere has been investigated. The analytical expressions for the average intensity and the degree of the polarization of a partially coherent hollow vortex Gaussian beam through a paraxial ABCD optical system are derived in turbulent atmosphere, respectively. The average intensity distribution and the degree of the polarization of a partially coherent hollow vortex Gaussian beam in turbulent atmosphere are numerically demonstrated. The influences of the beam parameters, the topological charge, the transverse coherent lengths, and the structure constant of the atmospheric turbulence on the propagation of a partially coherent hollow vortex Gaussian beam in turbulent atmosphere are also examined in detail. This research is beneficial to the practical applications in free-space optical communications and the remote sensing of the dark hollow beams.

A modified direct contact test was used to evaluate the antibacterial properties of four commercially available dental luting agents (RelyX Unicem, Ketac Cem, Ceramir Crown & Bridge and Harvard Cement) and two reference materials (glass-ionomer cement and calcium aluminate cement) compared to a negative-control material (PMMA). Streptococcus mutans bacteria were placed in direct contact with specimens that had been aged for 10 min, 1 day, and 7 days, in order to test the antibacterial properties of the materials. A metabolic assay containing resazurin was used to quantify the amount of viable bacteria remaining after the direct contact tests. The effects of pH and fluoride on bacteria proliferation were also evaluated. Strongest antibacterial properties were found for calcium aluminate cement, followed by Ceramir Crown & Bridge and RelyX Unicem. Ketac Cem, Harvard Cement, and the reference glass-ionomer cement showed bacteria content either higher than or not significantly different from the PMMA control in all instances. pH levels below 6.3 and above 9.0 were found to have negative effects on bacterial proliferation. No correlation between either acidic materials or fluoride release and antibacterial properties could be seen; rather, basic materials showed stronger antibacterial properties.

Person perception is a dynamic, evolving process. Because other people are an endless source of social information, people need to update their impressions of others based upon new information. We devised an fMRI study to identify brain regions involved in updating impressions. Participants saw faces paired with valenced behavioral information and were asked to form impressions of these individuals. Each face was seen five times in a row, each time with a different behavioral description. Critically, for half of the faces the behaviors were evaluatively consistent, while for the other half they were inconsistent. In line with prior work, dorsomedial prefrontal cortex (dmPFC) was associated with forming impressions of individuals based on behavioral information. More importantly, a whole-brain analysis revealed a network of other regions associated with updating impressions of individuals who exhibited evaluatively inconsistent behaviors, including rostrolateral PFC, superior temporal sulcus, right inferior parietal lobule, and posterior cingulate cortex.
