Microvesicles (or MVs) are plasma membrane-derived vesicles released from most eukaryotic cells constitutively during early apoptosis or at higher levels after chemical or physical stress conditions. This review looks at some of the functions of MVs in terms of intercellular communication and ensuant signal transduction, including the transport of proteins (unconventional protein export) as well as of mRNA and microRNA. MVs also have roles in membrane repair, the removal of misfolded proteins, and in the control of apoptosis. We also discuss the role MVs have been shown to have in invasive growth and metastasis as well as in hypoxia in tumours and cerebral ischaemia. The association of MVs in infectious and autoimmune disease is also summarised together with their possible use as therapeutic agents.

The methods of Plasma Membrane-derived Vesicle (PMV) isolation and quantification vary considerably in the literature and a new standard needs to be defined. This study describes a novel filtration method to isolate PMVs in plasma, which avoids high speed centrifugation, and to quantify them using a Becton Dickinson (BD) FACS Calibur™ flow cytometer, as annexin V-positive vesicles, larger than 0.2 μm in diameter. Essentially microvesicles (which comprise a mixture of PMVs and exosomes) from citrate plasma were sonicated to break up clumped exosomes, and filtered using Millipore 0.1 μm pore size Hydrophilic Durapore membranes in Swinnex 13 mm filter holders. Phosphatidylserine-positive PMVs detected with annexin V-PE were quantified using combined labelling and gating strategies in conjunction with Polysciences Polybead Microspheres (0.2 μm) and BDTrucount tubes. The PMV absolute count was calculated on the analysis template using the Trucount tube lot number information and expressed in PMV count/ml. Having estimated a normal reference range (0.51×10(5)-2.82×10(5) PMVs/ml) from a small sample of human donors, using the developed method, the effect of certain variables was investigated. Variations such as freezing of samples and gender status did not significantly alter the PMV absolute count, and with age plasma PMV levels were only marginally reduced. Smokers appeared to have reduced PMV levels. Nicotine, as for calpeptin was shown to dose-dependently (from 10 up to 50 μM) reduce levels of early apoptosis in THP-1 monocytes and to decrease the level of PMV release. Fasting individuals had 2-3 fold higher PMV absolute counts compared to non-fasting subjects.

Plasma membrane-derived vesicles (PMVs) also known as microparticles, are small membrane-bound vesicles released from the cell membrane via blebbing and shedding. PMVs have been linked with various physiological functions as well as pathological conditions such as inflammation, autoimmune disease and cardiovascular disease. PMVs are characterised by the expression of phosphatidylserine (PS) on the plasma membrane. PS, also expressed on apoptotic cells (ACs) enables macrophages to phagocytose ACs. As it is widely known that PMV production is increased during apoptosis, we were able to show that PMVs could compete dose dependently with ACs for the PS receptor on macrophages, so reducing phagocytosis of ACs. In a clinical setting this may result in secondary necrosis and further pathological conditions. In SLE in which there are raised PMV levels, there is an anti-phospholipid-mediated increase in PMV release, which can be abrogated by depletion of IgG. Our work provides an insight into how PMVs may play a role in the aetiology of autoimmune disease, in particular SLE.

Elderly men are an understudied population in the area of abuse and neglect. The typical elder abuse or neglect victim is described as a frail woman. The National Elder Abuse Incidence Study shows men to be victims as well and at higher risk than women for abandonment, the most extreme form of neglect. A qualitative study of older widowed men did not specifically explore issues of abuse and neglect, but found several of the men experiencing alienation from one or more of their adult children. This paper explores the dynamics of these "fractured relationships" and finds several patterns that may increase an older man's risk of being neglected when his needs for support increase. Many men are not functioning in a kinkeeper role and are not attempting to repair fractured relationships. Minimizing their own feelings may prevent men from understanding the extent of alienation felt by an adult child. doi:10.1300/J084v19n01_06.

Many faculty want to involve students more actively in laboratories and in experimental design. However, just "turning them loose in the lab" is time-consuming and can be frustrating for both students and faculty. We describe three different ways of providing structures for labs that require students to design their own experiments but guide the choices. One approach emphasizes invertebrate preparations and classic techniques that students can learn fairly easily. Students must read relevant primary literature and learn each technique in one week, and then design and carry out their own experiments in the next week. Another approach provides a "design framework" for the experiments so that all students are using the same technique and the same statistical comparisons, whereas their experimental questions differ widely. The third approach involves assigning the questions or problems but challenging students to design good protocols to answer these questions. In each case, there is a mixture of structure and freedom that works for the level of the students, the resources available, and our particular aims.

OBJECTIVE:: To examine the experiences of communication between older people and health and social care providers in six European countries. METHODS:: Focus groups were carried out with groups of older people (91 focus groups, 391 participants), and health and social care professionals (85 focus groups, 424 participants), in order to gain insights into concepts of good communications. Data collection and analysis continued concurrently according to the method of constant comparison. RESULTS:: Different styles of communication between professionals and older people were found to be capable of enhancing or jeopardising dignity. The use of appropriate forms of address, listening, giving people choice, including them, respecting their need for privacy and politeness, and making them feel valued emerged as significant ways to maintain older peoples' sense of self-worth and dignity. Despite being aware of good communication practices, health and social care professionals often failed to implement them. Lack of time, staff, resource scarcity, regulation and bureaucracy were cited as barriers, as was a lack of awareness and effort. CONCLUSIONS AND PRACTICE IMPLICATIONS:: The findings have important implications for health and social care professionals when they engage with older people.

The genetic architecture of floral traits involved in the evolution of self-pollination provides a window into past processes of mating system divergence. In this study, we use two generations of crosses between highly selfing and predominantly outcrossing populations of Arenaria uniflora (Caryophyllaceae) to determine the minimum number, average dominance relationships, and pleiotropic effects of genetic factors involved in floral divergence. Comparison of the F1 and F2 phenotypic means with the expectations of a completely additive model of gene action revealed a primarily additive genetic basis for floral characters associated with mating system variation. The exception was flower life span, which showed partial dominance of the outcrosser phenology. In contrast to similarly divergent species, the substantial differences in flower size between these A. uniflora populations appear to involve relatively few genes of large effect (minimum number of effective factors = 2.2 +/- 2.8 SE). In addition, correlations among traits in the F2 generation indicate that pleiotropy may be an important feature of the genetic architecture of floral evolution in A. uniflora. The evolution of selfing via major modifiers of floral morphology is consistent with other evidence for ecological selection for preemptive self-pollination in A. uniflora. Analyses of the genetic basis of autonomous selfing were complicated by hybrid breakdown in both F1 and F2 generations. Only F1 hybrids showed reductions in female fertility, but about 30% of F1 and F2 hybrids exhibited partial or complete male sterility. Male sterile flowers were characterized by short stamens, reduced petals, and a lack of protandry, as well as indehiscent anthers. This morphological breakdown mimics environmental disruptions of floral development and may result from novel genic interactions in hybrids.

The Gas-Grain Simulation Facility (GGSF) is a multidisciplinary experiment laboratory being developed by NASA at Ames Research Center for delivery to Space Station Freedom in 1998. This facility will employ the low-gravity environment of the Space Station to enable aerosol experiments of much longer duration than is possible in any ground-based laboratory. Studies of fractal aggregates that are impossible to sustain on Earth will also be enabled. Three research areas within exobiology that will benefit from the GGSF are described here. An analysis of the needs of this research and of other suggested experiments has produced a list of science requirements which the facility design must accommodate. A GGSF design concept developed in the first stage of flight hardware development to meet these requirements is also described.
