Summary With 20 figures and 5 tables Although yaks play an important role as companion or pack and draught animals on Chinese plateaus in alpine and sub-alpine regions, morphological studies and anatomical data on the lung of yak are sparse. To provide anatomical descriptions and morphometric data, 10 one-day-old yaks were examined by means of dissection, light and electron microscopy. The measurements of lung were made on dissected specimens and histological sections. Unequal dichotomous branching was demonstrated in the dissected tracheobronchial tree. The diameters of bronchial airways and height of epithelium were measured, and showed that the variation of diameters in airways was always greater than that of height of epithelium. In addition, the thickness of muscularis, cartilage and adventitia was examined, as well as the number of goblet and Clara cells in airways. Ultrastructural studies showed that the surface epithelium was mainly composed of goblet, ciliated, Clara and basal cells, similar to that in other domestic animals. Under electron microscope, two distinctive types of ciliated cells could be seen in the tracheobronchial epithelium. The first type contained some mitochondria, distended smooth endoplasmic reticulum (SER), little rough endoplasmic reticulum (RER) and numerous vacuoles in electron-lucent cytoplasm. The second type had dense cytoplasm with abundant mitochondria, RER and no vacuoles. Both types were rich of glycogen granules. The goblet cells in neonatal yak lung had the following characteristic features: dentate nucleus in dense cytoplasm, with stacks of RER and numerous dense membrane-bounded mucous droplets, which were round or oval, often with an electron-lucent core. The droplets were not confluent. Glycogen granules were numerous, and Golgi complex was occasionally present. Clara cells were dome-shaped and usually protruded into the airway lumen. Large amounts of SER and many secretory droplets were found within the cytoplasm. Several typical 'clefts' were also found in the cytoplasm.

Raman spectroscopy is a powerful tool for studying biochemical changes in the human body. We describe a miniature, confocal fibre optic probe intended to fit within the instrument channel of a standard medical endoscope. This probe has been optimized for the study of the carcinogenesis process of oesophageal malignancy. The optical design and fabrication of this probe is described including the anisotropic wet etching technique used to make silicon motherboards and jigs. Example spectra of PTFE reference samples are shown. Spectra with acquisition times as low as 2 s from resected oesophageal tissue are presented showing identifiable biochemical changes from various pathologies.

Abstract Streptococcus pyogenes are highly prevalent bacterial pathogens, especially in school-aged children. However, the characteristics of asymptomatic carriers vary geographically in different countries. We aimed to investigate S. pyogenes isolated from healthy schoolchildren in China. From 2007 to 2008, a total of 94 S. pyogenes isolates were obtained from healthy schoolchildren in Beijing and Chongqing, China. Antimicrobial susceptibility testing, determination of macrolide resistance genes (ermB, ermA and mefA), emm genotyping, pulsed-field gel electrophoresis (PFGE), and multilocus sequence typing (MLST) were performed. The resistance rate to macrolides was 96.8% and to tetracycline was 92.6%. All macrolide-resistant isolates exhibited constitutive resistance; 77 isolates (84.6%) had the ermB gene, while 14 isolates (15.4%) had the ermA gene. Among the macrolide-resistant isolates, the most frequent emm type was emm12 (51.6%), followed by emm22 (14.3%) and ST1815 (8.8%). PFGE analysis revealed 12 different patterns, with a given pattern having the same sequence type (ST) by MLST and the same emm type. In conclusion, the rate of macrolide resistance to S. pyogenes is currently very high in China. This is due primarily to the dissemination of a limited number of clones.

There is some controversy regarding the effects of HNO(3) on films of single-walled carbon nanotubes (SWCNTs). In this study we examined the change in sheet resistance of an HNO(3)-modified SWCNT film after different drying times at 85 degrees C using various analytical techniques. The shift and suppression in the Raman spectra, bleaching of the transition peaks related to van Hove singularities and a shift in the original peak in the C 1s XPS spectra provided evidence for p-type doping. A decrease in sheet resistance was also observed in the SWCNTs films due to the removal of residual N-methylpyrrolidone solvent on the surface and bundle of SWCNTs. These results suggest that p-type doping has a larger effect on the sheet resistance than the removal of residual N-methylpyrrolidone by an HNO(3) treatment.

OBJECTIVE. To review the pattern of ruptured intracranial aneurysms in terms of size, location, and the prevalence of multiple aneurysms in the Hong Kong Chinese population with subarachnoid haemorrhage. DESIGN. Retrospective study. SETTING. Three public hospitals in Hong Kong. PATIENTS. A total of 267 Chinese patients with subarachnoid haemorrhage from ruptured intracranial aneurysms between July 1998 and June 2002 were reviewed retrospectively. RESULTS. The patients had a mean age of 59 (range, 13-96) years, with a female-to-male ratio of 2:1. Concerning the age at presentation, males presented with ruptured intracranial aneurysms at a younger age (P=0.001) than females. Ruptured aneurysms were more commonly located in the anterior than posterior circulation (84% vs 16%). The posterior communicating artery (26%) and anterior communicating artery (22%) were the most common sites of rupture. As a whole, 64% of the aneurysms had a size of 5 mm or less. The anterior communicating artery had a higher proportion with a size of 5 mm or less compared to other locations (P<0.05). In this cohort, the prevalence of multiple aneurysms was 17%. There was no significant difference in the prevalence of multiple aneurysms between men and women (P=0.30). In patients with multiple aneurysms, the sizes of ruptured aneurysms were greater than those of the largest unruptured aneurysms (P<0.001). When compared with the group with single aneurysms, patients with multiple aneurysms had a smaller proportion of small aneurysms, sized 5 mm or less (P<0.05). CONCLUSIONS. The pattern of ruptured intracranial aneurysms in the Hong Kong Chinese population was different from western and Japanese populations. Although the distribution of locations for ruptured aneurysms was similar, Hong Kong Chinese had a larger proportion of small aneurysms sized 5 mm or less. The prevalence of multiple aneurysms in Hong Kong is comparable to that in the Japanese population, but lower than that in the western populations.

APRIL (A proliferation-inducing ligand) is a newly-identified member of the tumor necrosis factor family that induces pleiotropic biological responses, including immunological responses, IgA class switch and cell growth. It is associated with multiple diseases such as cancer and autoimmune diseases. High levels of APRIL mRNA can be detected in transformed cell lines and several malignant tumors; heparin sulfate proteoglycans (HSPG) are also involved in the APRIL tumor cell proliferation induction response. The interaction of APRIL and HSPG occurs through an N-terminal basic region on APRIL. We successfully expressed recombinant APRIL using an auto-induction system in E. coli. By using in situ cleavage, we generated the mature form of APRIL, comprising its N-terminal basic region, which has full biological activity: receptor binding capability and proliferation induction activity.

Abstract Objective: Radioimmunotherapy of cancer with radiolabeled antibodies has shown promise. We evaluated an anti-CD25 monoclonal Antibody, 7G7/B6, armed with (90)Y as a potential radioimmunotherapeutic agent for CD25-expressing lymphomas. Materials and Methods: The lymphoma model was established by subcutaneous injection of 1 x 10(7) SUDHL-1 cells into nude mice. The biodistribution of (111)In-7G7/B6 and therapeutic studies with (90)Y-7G7/B6 were performed in the tumor-bearing mice. Results: Therapy using (90)Y-7G7/B6 prolonged survival of the SUDHL-1 lymphoma-bearing mice significantly, as compared with either untreated mice or the mice treated with (90)Y-11F11, a radiolabeled isotype-matched control antibody (p < 0.001). All of the mice in the control and the (90)Y-11F11 treatment groups died by days 18 and 24, respectively. In contrast, 30% of the mice in the low-dose group (75 muCi of (90)Y-7G7/B6/mouse) and 75% in the high-dose group (150 muCi of (90)Y-7G7/B6/mouse) became tumor free and remained healthy for greater than 6 months. Conclusions: Our findings suggested that (90)Y-7G7/B6 is a potentially useful radioimmunotherapeutic agent for the treatment of patients with CD25-expressing lymphomas.
